Role of KatG in Francisella tularensis Susceptibility to Resazurin.

چکیده
The CDC classifies Francisella tularensis as a Category A bioterrorism agent. Due to the risk of potential release antibiotic-resistant F. strains, new therapeutics against must be developed. Resazomycins are resazurin (Rz)-based compounds that exhibit antimicrobial activity and other gram-negative bacteria. mode action resazomycins is not understood, but targets antibiotic were identified in high throughput screen for Rz-resistant isolates. FTL_1504 gene was being mutated all (Rzr) strains sequenced. encodes catalase KatG which catalyzes decomposition hydrogen peroxide water oxygen. Moreover, has also been shown play an essential role different antibiotic, isonazid. Four coding mutations characterized katG included single base pair substitutions deletion with resulting introduction premature stop codon. All Rzr mutants tested had reduced compared wild-type LVS three four loss expression. Based on this data, we hypothesize plays susceptibility resazurin. To address this, generating mutant measuring its Rz via agar dilution assays. Investigation contribution may unveil unique oxidative stress response family antibiotics

The use of resazurin as a novel antimicrobial agent against Francisella tularensis
The highly infectious and deadly pathogen, Francisella tularensis, is classified by the CDC as a Category A bioterrorism agent. Inhalation of a single bacterium results in an acute pneumonia with a 30-60% mortality rate without treatment. Due to the prevalence of antibiotic resistance, there is a strong need for new types of antibacterial drugs. Resazurin is commonly used to measure bacterial a...
متن کاملLack of OxyR and KatG Results in Extreme Susceptibility of Francisella tularensis LVS to Oxidative Stress and Marked Attenuation In vivo
Francisella tularensis is an intracellular bacterium and as such is expected to encounter a continuous attack by reactive oxygen species (ROS) in its intracellular habitat and efficiently coping with oxidative stress is therefore essential for its survival. The oxidative stress response system of F. tularensis is complex and includes multiple antioxidant enzymes and pathways, including the tran...
متن کاملRole of Glycosylation/Deglycolysation Processes in Francisella tularensis Pathogenesis
Francisella tularensis is able to invade, survive and replicate inside a variety of cell types. However, in vivo F. tularensis preferentially enters host macrophages where it rapidly escapes to the cytosol to avoid phagosomal stresses and to multiply to high numbers. We previously showed that human monocyte infection by F. tularensis LVS triggered deglycosylation of the glutamine transporter SL...
